چکیده
Other names: DKFZp686K23225 HGNC (Hugo): RASSF6 Location: 4q13.3 Local order: Centromere-AFP-AFM-RASSF6-IL8Telomere. Note Brief overview : The RASSF family of tumour suppressor genes (TSG) encode Ras superfamily effector proteins that, amongst other functions, mediate some of the growth inhibitory functions of Ras proteins. Several members of this family are inactivated by promoter DNA hypermethylation in a broad range of cancers, and the RASSF6 gene may be a frequent target of epigenetic inactivation in leukaemias. The RASSF6 protein is involved in the regulation of apoptosis partly by controlling the function of the proapoptotic mammalian serine/threonine kinases 1 and 2 (MST1 and MST2) and modulator of apoptosis 1 (MOAP-1).

RASSF proteins
What are RASSF proteins? RASSF stands for Ras association domain family. This family represents a group of ten mammalian proteins that contain a Ras association (RA) domain (Figure 1A). The first member of this family was identified as a binding partner of the Ras oncoprotein and was named NORE1 (novel Ras effector 1; now called RASSF5). The hunt for tumor suppressor genes on chromosome 3p21.3,...
متن کاملThe N-terminal RASSF family: a new group of Ras-association-domain-containing proteins, with emerging links to cancer formation.
The RASSF (Ras-association domain family) has recently gained several new members and now contains ten proteins (RASSF1-10), several of which are potential tumour suppressors. The family can be split into two groups, the classical RASSF proteins (RASSF1-6) and the four recently added N-terminal RASSF proteins (RASSF7-10). The N-terminal RASSF proteins have a number of differences from the class...
متن کاملNovel type of Ras effector interaction established between tumour suppressor NORE1A and Ras switch II.
A class of putative Ras effectors called Ras association domain family (RASSF) represents non-enzymatic adaptors that were shown to be important in tumour suppression. RASSF5, a member of this family, exists in two splice variants known as NORE1A and RAPL. Both of them are involved in distinct cellular pathways triggered by Ras and Rap, respectively. Here we describe the crystal structure of Ra...
متن کاملPromoter Methylation Status of Ras-Association Domain Family Members in Pheochromocytoma
Pheochromocytomas (PCCs) are rare neuroendocrine tumors that arise from the medulla of the adrenal gland or the sympathetic ganglia and are characterized by the secretion of catecholamines. In 30-40% of patients, PCCs are genetically determined by susceptibility genes as various as RET, VHL, and NF1. We have analyzed the Ras-association domain family members (RASSFs) in PCCs regarding their ina...
متن کاملRASSF4/AD037 is a potential ras effector/tumor suppressor of the RASSF family.
Activated Ras proteins interact with a broad range of effector proteins to induce a diverse series of biological consequences. Although typically associated with enhanced growth and transformation, activated Ras may also induce growth antagonistic effects such as senescence or apoptosis. It is now apparent that some of the growth-inhibitory properties of Ras are mediated via the RASSF family of...
